Just to clarify, I only want to have a single caliber, however I am not looking for a single handgun. I intend to purchase at a minum a dedicated handgun for the house as well as CCW and perhaps a hiking/SHTF spare as well. This will be in addition to the bolt-actions, battle carbines, rimfire and shotguns I currently have.

OR... is my thinking on this flawed, and I shouldnt attempt to use a single caliber to fill all roles, should I just keep .40SW around the house, and 9mm on my CCW and 44Mag for hunting sidearm? I was trying to be efficient and only stockpile a single ammo and reloading setup. Maybe im asking to much from 1 caliber.

Well I'll go on a different route. Buy yourself two revolvers, and one rifle, with all three chambering .357magnum. Any of the three can shoot .38 long colt, .38 Special, or .357 magnum.
1. Rifle Marlin 1894c
2. Ruger GP100, S&W model 27/28 4"bbl, S&W model 686/586, S&W 66/19
3. Ruger SP101. I would list the little S&W J-frame's, but in my opinion they(even the all steel models) are to light for the cartridge.

You can also do the same with the .44 magnum/.44 special.

This.

Your question was what one handgun calibre would you have? If I only had one it would be a .22 lr. You didn't ask which one do you prefer for self defense?

If I could only have one caliber: probably .22lr

If I could only have one for self-defense: .45 ACP

Apparently the term "stopping power" is not popular here, but the reason it is a well known saying is because there is truth in it. Ask why the police went to .40 from 9mm, or the FBI to 10mm. Reason: you can STOP a target with less hits on the target. Sure a well placed 9mm could stop someone, or if not well placed, then a half-dozen of them pretty close may do, but I feel better with a caliber that has a proven track record of dropping the bad guy fast.

Personally, I think you are limiting yourself too much by trying to find a one caliber solution. I tried to not get too many calibers, but it just happens. Just stockpile ammo in the most common ones you shoot and have some rounds for the others and you will be fine.

Nope! The Reason was politics and false assumptions - knee jerk reaction to a tragedy (no different than what it is going on in 2A arena right now) - FBI wanted to "do something" about handguns sucking at killing. Add Glock's unparalleled marketing department - Glock managed to put a pistol on the market chambered in 40 Smith and Wesson ... before Smith and Wesson!!!

I am sure this has been beaten to death, but the only way to STOP a target is to disrupt central nervous system; and since 22LR has a fantastic record of one-shot stop/kills, your .356 vs .400 argument is irrelevant.

This is some very good advice. Take a 9mm to a pumpkin some time. When you have sufficiently put a few holes in it, switch to 357 and behold the carnage.
