You're going to want to purify the crap out of it if you're planning to drink it - REGARDLESS of roof type.. so go for it.
If you plan to just use it to watering the garden and stuff like that.. no need to worry about it at all. no diff than regular runoff in your yard.

get a T/Y adapter on your downspout so you have a trash catch at the bottom - or a straight pipe if you need to do that. then connect your catch barrel to the higher diverted line. that way you can keep trash out of your catch barrels a bit better.

Back when we had open top tanks for our houses and cisterns we had a column of charcoal that it ran through before it went to the tank. But that was for drinking. Also there was more cholera and typhus around then too!
OSU has a plan with a filter that purifies pond water clean enough for drinking. Activated charcoal is part of it.
A former co-worker had to do that when their well failed.

Look around locally for overpack drums. They're used by shipping companies for leaky 55 gal drums. They clean up nicely, hold way over 55 gallons, and have a huge screw off lid for cleaning. No need to filter sediment first, just an annual cleaning at the end of the season.
